Influence of Certain Cations on Activity of Succinyl CoA Synthetase From Tobacco.
نویسنده
چکیده
Succinyl CoA synthetase from Nicotiana tabacum exhibited a requirement for univalent and divalent cations. Mn(2+) replaced Mg(2+) in the assay medium and Co(2+) and Ca(2+) partially replaced Mg(2+). Addition of Zn(2+) resulted in no enzyme activity. The enzyme was activated by univalent cations K(+), Rb(+), NH(4) (+), and Na(+); Li(+) showed little or no activation. Maximum enzyme activity varied significantly with potassium salts of different anions. Greatest activation was obtained with K(3)PO(4) and, respectively, KCl, KNO(3), K(2)SO(4) and KF exhibited steadily decreasing enzyme activation.

Soluble -aminolevulinic acid synthetase of rat liver. II. Studies related to the mechanism of enzyme action and hemin inhibition.
The present study describes some characteristics of soluble hepatic d-aminolevulinic acid synthetase, including end product inhibition by hemin. The enzyme is shown to contain a -SH group at its active center which is associated with the initial binding of the cofactor pyridoxal-S/-PO4 to the enzyme. In addition, the -SH group may influence the activation of d-aminolevulinic acid synthetase by ...

A succinyl coenzyme A synthetase preparation from Escherichia coli has the following characteristics: apparent chromatographic, sedimentation, and electrophoretic homogeneity; molecular weight of about 141,000; Azso (1 mg per ml) = 0.511; higher specific activity than previous preparations; and identity by several criteria with a phosphorylated protein (E-P) obtained upon exposure to Mgf+, inor...

The sucCD gene of Advenella mimigardefordensis strain DPN7 encodes a succinyl coenzyme A (succinyl-CoA) synthetase homologue (EC 6.2.1.4 or EC 6.2.1.5) that recognizes, in addition to succinate, the structural analogues 3-sulfinopropionate (3SP) and itaconate as substrates. Accumulation of 3SP during 3,3 -dithiodipropionic acid (DTDP) degradation was observed in Tn5::mob-induced mutants of A. m...
متن کاملX-linked sideroblastic anemia due to carboxyl-terminal ALAS2 mutations that cause loss of binding to the β-subunit of succinyl-CoA synthetase (SUCLA2).
Mutations in the erythroid-specific aminolevulinic acid synthase gene (ALAS2) cause X-linked sideroblastic anemia (XLSA) by reducing mitochondrial enzymatic activity. Surprisingly, a patient with the classic XLSA phenotype had a novel exon 11 mutation encoding a recombinant enzyme (p.Met567Val) with normal activity, kinetics, and stability. Similarly, both an expressed adjacent XLSA mutation, p...
متن کاملStudies of the acetyl coenzyme A synthetase reaction. IV. The requirement for monovalent cations.
Acetyl coenzyme A synthetase, which catalyzes the biosynthesis of acetyl coenzyme A from acetat,e, adenosine triphosphate, and coenzyme A, has previously been obtained in highly purified form from bovine heart mitochondria (1). The enzyme appeared to be fairly homogeneous because only a single protein peak was detected in two chromatographic systems and the pattern observed in the analytical ul...
